چکیده

Membrane Transfer Process for the Creation of Low-Noise Solid State Nanopore Devices Tarun Jain Nanopore sensors are an emerging technology whereby the modulation of a current trace upon passage of the analyte through the nanopore is used to infer the properties of the analyte. This technology is particularly attractive because it is label-free, rapid, and intrinsically single molecule. However, the ability to infer molecular properties with nanometer precision, either in size or sequencing, is obscured by noise in the measurement. The precision of this inference can be significantly improved by reducing noise and by performing multiple measurements on the same molecule. Solid state nanopores, made in free standing membranes, however, have traditionally exhibited high levels of capacitive noise at 100 kHz bandwidths, as well as poor confinement of the electric field around the nanopore. In this thesis, a novel device concept is designed for creating solid state nanopores whereby the free standing membrane is transferred over a PDMS microchannel. By eliminating the silicon wafer backing, capacitive noise is dramatically reduced. Furthermore, the microchannel confines the electric field outside the nanopore, thereby enhancing the ability to perform multiple measurements.

Integration of solid-state nanopores in a 0.5 μm CMOS foundry process.

High-bandwidth and low-noise nanopore sensor and detection electronics are crucial in achieving single-DNA-base resolution. A potential way to accomplish this goal is to integrate solid-state nanopores within a CMOS platform, in close proximity to the biasing electrodes and custom-designed amplifier electronics. Here we report the integration of solid-state nanopore devices in a commercial comp...

Fine-tuning the Size and Minimizing the Noise of Solid-state Nanopores

Solid-state nanopores have emerged as a versatile tool for the characterization of single biomolecules such as nucleic acids and proteins. However, the creation of a nanopore in a thin insulating membrane remains challenging. Fabrication methods involving specialized focused electron beam systems can produce well-defined nanopores, but yield of reliable and low-noise nanopores in commercially a...

Prevention of Dielectric Breakdown of Nanopore Membranes by Charge Neutralization

To achieve DNA sequencing using a solid-state nanopore, it is necessary to reduce the electric noise current. The noise current can be decreased by reducing the capacitance (C) of the nanopore device. However, we found that an electric-charge difference (ΔQ) between the electrolyte in one chamber and the electrolyte in another chamber occurred. For low capacitance devices, this electric-charge ...

DNA translocation through graphene nanopores.

We report on DNA translocations through nanopores created in graphene membranes. Devices consist of 1-5 nm thick graphene membranes with electron-beam sculpted nanopores from 5 to 10 nm in diameter. Due to the thin nature of the graphene membranes, we observe larger blocked currents than for traditional solid-state nanopores. However, ionic current noise levels are several orders of magnitude l...

Solid-state nanopores and nanopore arrays optimized for optical detection.

While conventional solid-state nanopore measurements utilize ionic current, there is a growing interest in alternative sensing paradigms, including optical detection. However, a limiting factor in the application of optical schemes in particular is the inherent background fluorescence created by the solid-state membrane itself, which can interfere with the desired signal and place restrictions ...
